I found on cheapoair.com a round trip flight to Berlin leaving 6 May and returning 28 May for $515 + tax ($736 total). The flight to Frankfurt is on United Airlines, and the flight Berlin is Lufthansa. On the returning flight, (to Frankfurt, then back to Detroit) both use Lufthansa.
Should I book this?

I saw your itinerary in the other section of the forum. You may want to consider booking open-jaw tickets if the price difference isn't too great. Also, a quick googling of Cheapoair reveals that their reputation concerning customer service is "mixed."

That's not really too bad - I've seen flights from Detroit to Frankfurt for 300-400 dollars roundtrip according to airfarewatchdog.com but they only go up to April as for now.

That is quite a bit cheaper than what the United.com web site has.
I would play around with it a bit and see if you can get another city to return to.
Even if you couldn't get Amsterdam, for example, maybe you could get Cologne, and it's much less of a trek from Amsterdam to Cologne (or Dusseldorf even better) than it is back to Berlin. The German cities seem to be the cheapest places to fly into/out of, right now.
Check to see when cheapoair.com confirms the tickets. If it's like many other consolidators, it sells you the ticket, but does not actually confirm it for 48 hours or so. That's different than a site like Orbitz where you get instant confirmation (or as you can do on the airline's own web site). This is not a big deal, but you just want to be absolutely sure your tickets exist before proceeding.
Once you know you have your tickets, you can phone up United and get a seat selection (this will prove that United knows about your ticket).
