I will be flying out of Rome late in the evening, which means I will have the entire day to spend in Rome after checking out of my hotel. I would rather not carry around my luggage the entire day, but I will if I have to.
Does anyone have any suggestions on what I do? Should I ask the hotel if they will hold onto my pack for me until the end of the day?

Yes, that's the best solution. Most hotels and hostels will do this, but not all B&Bs, so just check ahead of time to ensure your place does.

I do think it depends on the hotel or hostel. Don't be dismayed if they turn you down. You can always find a train station (they often have lockers) or a luggage storage place and pay a small fee (usually no more than a few euros) for your bags.
I find that asking ahead of time (perhaps when you check in) is the best way because they'd appreciate your upfrontness rather than springing it on them last minute as you check out.

There is always lockers at train stations and airports too if you're hotel won't accommodate you? Depending on how your departing, it might be a good idea to first drop off your luggage and then boot around town - one less thing to worry about! Having to lug around luggage is definitely on of my most unpleasant aspects of traveling. Good luck and have fun!
